Unlike other Phospholipase C (PLC) isozymes, PLC beta enzymes contain a
region of sequence homology of ~250 amino acids near the C-terminus.
This region is involved in activation of the lipase activity of PLC beta
by GTP-bound G alpha q subunits and in turn acts as a GTPase Activating
Protein (GAP) for G alpha q. This C-terminal 250 amino acid domain contains
no obvious homology to known structures. I have solved the
structure of this C-terminal domain representing amino acids
880-1154 of a PLC beta isolated from turkey erythrocytes.
Initially crystals from this construct diffracted poorly (~7 A),
so a region spanning amino acids 946-978 was deleted. This
region is highly variable among PLCs, is protease-sensitive
and is a site for serine phosphorylation. The crystals were grown
between pH 5.7 to 6.3 and are triclinic. I would like to withhold
information about the oligomerization state at present as I
feel this is one of the interesting aspects of this structure.

9. Current state of the experimental work

This structure was solved by MAD using crystals containing
selenomethionine-labeled protein. I have refined to a resolution
of 2.4 A, with an R and Rfree of 25.4 and 29.3 respectively. Little
more will be done with regard to further refinement as there are no
outliers in Ramachandran phi/psi space and the geometry is well
within what is expected for a 2.4 A structure.
