7. Additional Information
In 1988 (1BKS; Hyde et al.) the three dimensional structure of the tryptophan synthase alapha2beta2 complex from Salmonella typhimurium has been determined by X-ray analysis. However, the structure of the alapha or beta2 subunit alone has not yet been determined. We picked up alapha-subunit from hyperthermophile, Pyrococcus furiosus
Because the sequence homologue of both proteins was 31.5 %, molecular replacement method (MR) using the structure of alapha-subunit in the alapha2beta2 complex of tryptophan synthase from S. typhimurium (1BKS) were tried using AMoRe. But all attempts to refine the solution failed. Therefore, the structure of alpha-subunit from P. furisus was solved by the method of multiple isomorphous replacement (MIR) using two derivatives; Pt and Hg.
Recently, we could determine the structure of alapha subunit from P. furiosus at a resolution of 2.0 A(free R = 24.9, R = 19.8).
