The structure of EH2 was determined after subcloning amino acids 121-218 from human Eps15 in a prsetA vector, and was expressed in E.coli.
The protein contains 5 N-terminal residues (DRWGS) and three C-terminal residues (ELI) from the expression vector.

About 30 EH domains in 20 proteins have been identified based on amino acid sequence.
A calcium binding site is located in the current structure. The location of this calcium binding site varies between EH domains.
